What doses did the studies use? They began with a small amount and added more in slow steps. That pace helped people bear the early stomach trouble.
Researchers gave the shot each week and the pill each day on an empty stomach. Every number below appears in a trial paper or on the approved label.

How the Semaglutide dose rose in shot studies
The shot enters the fatty layer beneath the skin, and studies raised the semaglutide dose in steps. For long-term weight care, it began at 0.25 mg through weeks 1-4, then rose to 0.5 mg, 1.0 mg, and 1.7 mg. STEP 1 [1] and SELECT [3] used the steady 2.4 mg dose.
Studies for type 2 diabetes used lower steady amounts. They began at 0.25 mg, then moved to 0.5 mg and 1.0 mg. SUSTAIN also studied a 2.0 mg choice.
The slow rise had a purpose. A review found stomach problems were usually mild to moderate and soon passed, with most trouble during the weeks when the amount increased [23]. These figures report the studies and aren't a personal dosing plan.
How Semaglutide dosage worked for the daily pill
Oral semaglutide dosage follows a different plan. An added ingredient moves a small share of medicine from the stomach into blood [18]. Food or a large drink can reduce that share. Scientists also changed the protein part they call 8, one item in its chain, so the medicine lasts longer.
The diabetes studies began with 3 mg each day for 30 days, then raised the amount to 7 mg and 14 mg. People took the tablet before eating, drinking, or taking other pills. They waited about 30 minutes and used no more than 120 mL of water, less than a small drinking glass [18].
Other studies tested 25 mg and 50 mg each day. Those pills lowered sugar readings and body weight more than the 14 mg pill [11]. These are study figures, not directions for you.
Where the Semaglutide injection goes
A semaglutide injection enters the fatty layer just under the skin, not a vein or muscle. SUSTAIN, STEP, and SELECT all used this weekly shot [1][2][3].
The approved label usually says to refrigerate unused pens. A pen in use may be kept out of the refrigerator, but no warmer than 30 degrees celsius, a very warm room, for up to 56 days. The exact limit depends on the pen. A product mixed by a pharmacy may carry a limit of 28 days and different storage rules.
The approved label and study papers supply these facts; they aren't directions for the pen in your hand. Check its own label, and ask a pharmacist if the storage rule isn't clear.
How long Semaglutide stays in the body
Half of semaglutide leaves the blood in about one week. The timing is much the same for the shot and tablet; by about the fifth week after the last dose, nearly all has left [17].
The medicine rides on a common blood protein. Scientists also changed the protein part they call 8, so a body enzyme called DPP-4 clears the drug more slowly. Together, those changes keep semaglutide in the blood for days.
That long stay lets one shot cover a week, and it is also why the pregnancy label names a delay [17]. Studies checked metformin for diabetes, warfarin to thin blood, atorvastatin for cholesterol, and digoxin for the heart. Semaglutide didn't change their blood levels enough to matter [12]. Even so, your prescriber needs a complete list of everything you take.
